Spider-Man follows Peter Parker, an orphaned high schooler who pines after popular girl-next-door Mary Jane Watson (Kirsten Dunst). While on a science class field trip, Peter is bitten by a genetically-engineered "super spider." As a result, Peter gains superhuman abilities, including increased strength, speed, and the abilities to scale walls and generate organic webbing. After his beloved Uncle Ben (Cliff Robertson) is murdered, the teenager realizes that he must use his newfound abilities to protect New York City. Meanwhile, wealthy industrialist Norman Osborn (Willem Dafoe), the father of Peter's best friend Harry Osborn (James Franco), subjects himself to an experimental performance-enhancing serum, which creates a psychotic and murderous split personality. Donning a military battlesuit, Norman becomes a freakish "Green Goblin", who begins to terrorize the city. Peter, as Spider-Man, now must do battle with the Goblin, all while trying to express his true feelings for Mary Jane.
Peter Parker, secretly the superhero Spider-Man, is estranged from both love interest Mary Jane "M.J." Watson and best friend Harry Osborn, and additionally discovers his Aunt May is facing eviction. He also finds himself suffering temporary, but recurring losses of his powers, often in life-threatening situations. Harry, now head of Oscorp's research division, is sponsoring a fusion power project by nuclear scientist Otto Octavius', who befriends and mentors Peter. While handling hazardous materials, Octavius wears a harness of powerful robotic tentacle arms with artificial intelligence. During a public demonstration that Peter and Harry attend, a power spike causes the fusion reactor to destabilize. Octavius refuses to shut down the reactor, which goes critical
Spider-Man 3 picks up one year after the events of the second film. Peter is still seeing Mary Jane Watson, while Harry Osborn succeeds his father as the new Green Goblin. Eddie Brock, who like Peter is a photographer for the Daily Bugle, sets out to defame Spider-Man and incriminate him. Flint Marko, an escaped convict, falls into a particle accelerator and becomes a shape-shifting sand monster later known as Sandman. Peter later learns that Marko is the one that killed Uncle Ben, causing Peter's own dark intentions to grow. This vendetta is enhanced by the appearance of the mysterious black alien symbiotic substance that bonds to Peter, resulting in the formation of a new black costume. Once Peter separates himself from the alien, it finds a new host in the form of Brock, resulting in the creation of Venom.
A young Peter Parker discovers that his father Richard Parker's study has been burgled. Peter's parents gather hidden documents, take Peter to the home of his Aunt May and Uncle Ben, and then mysteriously depart. Years later, a teenage Peter attends Midtown Science High School, where he is bullied by Flash Thompson, and has caught the eye of Gwen Stacy. At home, Peter finds his father's papers, and learns his father worked with fellow scientist Dr. Curt Connors at Oscorp in the field of cross-species genetics. Sneaking into Oscorp, Peter enters a lab where a "biocable" is under development from genetically modified spiders, one of which bites him. He later discovers he has developed spider-like abilities, such as sharp senses, reflexes and speed.
OsCorp scientist Richard Parker records a video message to explain his disappearance. Later, he and his wife, Mary, are aboard a private jet hijacked by an assassin sent to kill Richard and his wife. Richard and Mary fight off the man, but the ensuing struggle causes the plane to crash, killing both Richard and Mary, after he uploads the video. In the present time, Richard's son, Peter, continues to fight crime as Spider-Man. He pursues and apprehends Aleksei Sytsevich. Later, Peter meets with his girlfriend Gwen Stacy at their high school graduation ceremony and, insisting he keep his vow to her late father, ends their relationship. Peter's childhood friend, Harry Osborn, returns to Manhattan to see his terminally-ill father, Norman, CEO of OsCorp. Norman explains his illness is genetic, and Harry is at the age where it first develops. Norman gives Harry a small device he claims contains his life's work. The next day, Norman dies, and Harry is appointed the new OsCorp CEO.

Following the Battle of New York, Adrian Toomes and his salvage company are contracted to clean up the city, but their operation is taken over by the Department of Damage Control (D.O.D.C.), a partnership between Tony Stark and the U.S. government. Enraged at being driven out of business, Toomes persuades his employees to keep the Chitauri technology they have already scavenged and use it to create and sell advanced weapons. Eight years later, Peter Parker is drafted into the Avengers by Stark to help with an internal dispute, but resumes his studies at the Midtown School of Science and Technology when Stark tells him he is not yet ready to become a full Avenger.
